EFFECTS OF CHLOROGENIC ACIDS IN RAT FIBROBLAST AND NEUROGLIAL CELLS TREATED WITH CADMIUM

Abstract
-Abstract-
This study was carried out to develop compounds repairing rat fibroblast and
neuroglial cells damaged by cadmium in vitro.
The experimental groups devided into sex groups in chlorogenic acid : control groups
was cultured in the media only, NR50 and MTT 50 groups
were cultured in the media containing NR50 of cadmium and four
experimental groups were cultured in the media containing NR50 of
cadmium and various concentrations of chlorogenic acid.
The 5.0¡¿104 cell/§¢ of rat fibroblasts and neuroglial cells in each well
of 24 multidish were cultured. After 24 hours, the cells were treated with solution of all
groups. After the fibroblast and neuroglial cells of all groups were cultured in same
condition for 48 hours, Neutral red (NR) and tetrazolium MTT (MTT) of the
colorimetric assay were performed to evaluate the cytotoxicity of cell organelles. Also,
the lactate dehydrogenase (LDH) activity was measured for membrance toxicity. The
light microscopic study was carried out to observe morphological changes of cultured rat
fibroblast and neuroglial cells.
The results were as follow :
1. According to the NR absorbance, the effects of 50 ¥ìM chlorogenic acids were active
in fibroblast and neuroglial cells. but 100 ¥ìM chlorogenic acids were decreased effects
again.
2. By means of MTT absorbance, the repaired effects of chlorogenic acid 50 ¥ìM
concentration were active in fibroblast and neuroglial cells. However, 100 ¥ìM
chlorogenic acids were decreased repaired effects again.
3. In LDH activity had good repaired effects in fibroblast. chlorogenic acid 50 ¥ìM
concentration had good repaired effects in neuroglial cells. but ¥ìM concentration were
decreased repaired effects again.
4. The cell numbers and cell shapes were repaired in the 25 ¥ìM and 50 ¥ìM
chlorogenic acid but 100 ¥ìM of showed degeneration again. These results indicated that
chlorogenic acid decreased the cytotoxicity of cadmium in cultured fibroblast and
neuroglial cells.
